Solid Media and Inoculation Techniques

• Solid media can be contained in test tubes or Petri dishes, with inoculation performed using the streaking technique, which can be either straight or wavy.

• Inclined solid media tubes are utilized for studying bacterial growth patterns, observing metabolic characteristics, or short-term preservation of microorganisms.

Straight Streak Technique

• The straight streak method is used for macroscopic characterization of cultures. It requires a sterile inoculating loop (azabacteriológica), a flame source (mechero), culture media in tubes, and bacterial or yeast cultures.

• After taking the inoculum with the sterile loop and making a straight line on the medium's surface, it is essential to sterilize the loop again after use.

Wavy Streak Technique

• The wavy streak method aims to achieve abundant microorganism development for short-term preservation. Similar materials as in the straight streak technique are used.

• The inoculum is distributed by creating closed wavy streaks across the entire surface of the medium after inserting the sterile loop into an inclined tube.

Combination Techniques for Metabolic Testing

• For certain metabolic tests, a combination of stab and streak techniques is employed. This involves using an inoculating needle instead of a loop.
